不同浓度TTC染色对高血压性脑卒中病灶面积的评估 被引量:4

摘要 【目的】探讨不同浓度2,3,5-氯化三苯基四氮唑(TTC)对高血压性脑卒中病灶的检出效能。【方法】将卒中易感型自发性高血压大鼠(SHR-SP)随机分为两组,一组用于比较两种不同浓度TTC染色结果(0.05%和2.0%);另一组用于比较0.05%TTC和HE染色方法计算出的脑卒中面积百分比及两者间的相关性。【结果】0.05%TTC组脑卒中面积与2.0%TTC组无显著性差异,且两组比较,计算后的脑卒中面积密切相关。0.05%TTC组与HE染色比较,脑卒中的面积百分比无显著性差异,且两组间脑卒中面积百分比具有高度相关性。【结论】TTC染色是一种比HE染色更为经济、快捷的检测实验性脑卒中范围的染色方法,并且0.05%TTC与常用浓度检出效能具有高度相关性,而其花费只为常规方法的1/40,是理想的TTC染色浓度。 [Objective] To investigate a specific procedure and optimized concentration of 2, 3, 5-triphenyhetrazolium chloride (TTC) staining for the detection and quantification the stroke lesion due to hypertension. [Method] Stroke- Prone Spontaneously Hypertensive Rats (SHR-SP) were randomly divided into 2 groups. Sets of two serial slices were set in 2.0% and 0.05% TTC individualy. For comparison with HE staining, the percent area of stroke lesions in each sections by above staining techniques were quantified by image analysis. [Results] When the stroke areas in juxtaposing faces of consecutive slices stained with different TTC concentrations were compared, there were no statistical difference between those by 0.05 % TTC and those by 2.0% TTC. Regression analysis demonstrated a very flood correlation between stroke areas revealed by the two TTC concentrations. The size of areas of stroke determined by 0.05 % TTC and HE staining was not significantly different, and presented a satisfied correlation. [Conclusion] As compared with HE staining, staining with TTC is a rapid, inexpensive, and reliable method for the detection and quantification of stroke in rats. Moreover, TTC at 0.05 % concentration is optimal for staining adult rat brain, which has the same effect to detect and quantify the stroke while reduces the costs to 1/40.
