产虾青素海洋酵母菌YS-185原生质体制备与紫外诱变 被引量:4

摘要 对海洋酵母菌YS-185原生质体制备、再生条件及紫外诱变育种进行了研究。结果显示,对数生长中后期菌龄较易形成原生质体,用EDTA和β-巯基乙醇混合溶液预处理15min有更高的原生质形成率,原生质体形成及再生率最佳条件为:0.5%蜗牛酶,35℃、酶解60min,对原生质体紫外线辐射60s诱变出YS-185-19突变株,其色素产量较出发菌株提高了93.4%。 The suitable condition for preparation and regeneration of marine protoplast was studied, and high yield astaxanthin producing strains were obtaine let mutagenesis. The protoplast formation was found to be more easily induced yeast YS-185 d by ultravioduring metaphase and anaphase of logarithmic growth; and higher rate of protoplast formation was obtained when treated by the mixture of β-mercaptoethanol and EDTA for 15 min. Optimum condition for protoplast formation and regeneration was 0.5 G snaillase treatment for 60 min at 35 ℃. A high astaxanthin yield mutant strain of YS-185-19 was obtained after ultraviolet mutagenesis for 60 s, for which the pigment content was 93.4% higher than the original strain.
