摘要 目的探讨低氧诱导因子HIF-1α和HIF-2α在人肾透明细胞癌组织中的表达及其临床意义。方法应用RT-PCR的方法从转录水平检测了56例原发性散发肾细胞癌和52例正常肾组织中HIF-1αmRNA及HIF-2αmRNA的表达情况,应用Western-Blot方法从蛋白质水平检测了HIF-1α和HIF-2α的表达情况。结果HIF-1αmRNA在肾透明细胞癌标本组织中的表达率为87.5%,较对照组织中增高,差异有统计学意义(P<0.05);HIF-2αmRNA在肾透明细胞癌标本组织中的表达率为94.6%,较对照组织中增高,差异有统计学意义(P<0.05)。56例CCRCC组织标本中52例表达HIF-1α蛋白,阳性率为92.9%,较对照组织中增高,差异有统计学意义(P<0.05)。56例CCRCC组织标本中54例表达HIF-2α蛋白,阳性率为96.4%,较对照组织中增高,差异有统计学意义(P<0.05)。结论HIF-1α和HIF-2α的表达可能对肾透明细胞癌的发生和发展有重要作用。 Objective To explore the expression and clinical significance of Hypoxia-inducible factors HIF-1α and HIF-2α,in clear cell renal cell carcinoma(CCRCC).Methods The expression of HIF-1α mRNA and HIF-2α mRNA was checked in 56 cases of primarily diverged renal carcinoma and 52 cases of normal renal tissues at transcription level by RT-PCR.The expression of HIF-1α and HIF-2α on the protein level was detected by Western-blot.Results The expression rate of HIF-1α mRNA in tissue specimen of CCRCC was 87.5%,increased more significantly than the control group(P〈0.05);expression rate of HIF-2α mRNA in tissue specimen of CCRCC was 94.6%,increased more significantly than the control group(P〈0.05).In the 56 CCRCC tissue specimens,52 cases expressed HIF-1α protein,the positive rate was 92.9%,increased more significantly than the control group(P〈0.05).In 56 casesed CCRCC tissue specimens,54 cases expressed HIF-2α,and the positive rate was 96.4%,increased more significantly than the control group(P〈0.05).Conclusions The expression of HIF-1α and HIF-2α on the mRNA and protein level possibly play an important role in the genesis and development of CCRCC.
