摘要
目的:建立一种高效快速的高效液相色谱-蒸发光散射检测法(HPLC-ELSD)测定中药当归中蔗糖含量的方法。方法:采用Shodex NH2P-50 4E(4.6mm×250mm,5μm)色谱柱,以乙腈-水(78:22)为流动相,流速:1.2mL·min^(-1),柱温:60℃,漂移管温度:90℃,气体流量:2.1SLPM。结果:蔗糖在40.32~403.2μg线性关系良好。结论:该法可以用于传统中药当归药材中蔗糖含量的检测。
Objective: To build up a simple and rapid method by HPLC with Evaporative Light Scattering Detection (ELSD) method for the determination of sucrose in Radix Angelicae Sinensis. Methods: The HPLC separation was performed on a Shodex NH2P-50 4E column using acetonitrile-water (78:22) as mobile phase at a flow rate of 1.2mL·min^- 1 The drift tube temperature of ELSD was set at 90℃ with the nitrogen flow rate of 2. 1SLMP. Results: The linear range was from 40.32 to 403.2 μg. Conclusion : The determination result by this method was satisfactory in separation and determination of sucrose in traditional chinese herb Angelicae sinensis.
