50例恶性神经胶质瘤中细胞周期蛋白D基因扩增检测分析 被引量:1

An Analysis of Amplification of CyclinD Genes in 50 Cases with Malignant Glioma

摘要 [目的]探讨人恶性神经胶质瘤中细胞周期蛋白D基因扩增的生物学意义。[方法]采用实时荧光定量PCR技术检测50例人恶性神经胶质瘤及对应正常脑组织中CCND1、CC-ND2、CCND3基因的扩增水平。[结果]50例胶质瘤标本有36例(72.0%)cyclinD1扩增、34例(68.0%)cyclinD3扩增、23例(46.0%)cyclinD2扩增,正常脑组织均无扩增。CCND基因扩增在胶质瘤与正常脑组织中差异有显著性(P<0.01),随着胶质瘤恶性程度升高,CCND1和CCND3基因扩增阳性率增高,且两者之间存在显著性正相关。[结论]人恶性神经胶质瘤中细胞周期蛋白D基因过度扩增,CCND1和CCND3随胶质瘤病理分级增加而升高,提示CCND基因的异常扩增在胶质瘤的发生与发展中起着重要的促进作用。
