摘要
[目的]研究华蟾素(Cino)对胃癌细胞的增殖抑制及诱导凋亡作用。[方法]Cino与胃癌细胞株BGC-823共同孵育不同时间后,应用荧光显微镜、MTT检测技术、流式细胞分析仪技术观察其对胃癌细胞株BGC-823的凋亡诱导作用。[结果]不同浓度Cino作用于胃癌细胞株BGC-823后,可看到典型的细胞凋亡形态学变化:细胞固缩,染色质凝集,呈新月形紧贴于核膜周边,核碎裂,凋亡小体形成等。MTT法显示Cino对人胃癌细胞有显著的增殖抑制作用,且有时间和浓度依赖性。Cino(10μg/ml)处理的BGC-823细胞48h,流式细胞术检测出现明显的亚二倍体凋亡峰,凋亡率可达25.1%。[结论]Cino对胃癌细胞具有诱导细胞凋亡的作用,细胞凋亡发生率呈剂量和时间依赖性,具有治疗胃癌的潜在价值。
[Purpose] To investigate effect of cinobufacin on inhibiting proliferation and inducting apoptosis in gastric carcinoma cell. [Methods] Apoptosis in gastric carcinoma cell line BGC-823 induced by cinobufacin was investigated with fluoroscope,MTT assay and flow cytometer. [Results] Gastric carcinoma cell line BGC-823 was incubated with cinobufacin in various concentration. Morphologic features of apoptosis,including cell shrinkage,nuclear condensation,DNA fragmentation and formation of apoptotic bodies were found. With MTT assay,Cino could remarkably inhibit proliferation of human gastric carcinoma cell line BGC-823 in a time-and concentration-dependent manner. An obvious subdiploid apoptosis peak in BGC-823 cells was exhibited with flow cytometer at 48 hours incubated with cinobufacin(10μg/ml).The rate of apoptosis was 25.1%. [Conclusion] Cinobufacin can induce apoptosis of gastric carcinoma cell with concentration-and time-dependence. It might have a promising prospect in the treatment for gastric carcinoma.
