摘要
将1日龄小鼠卵巢移植入成年雄鼠肾囊下,分别于移植后21,28和35 d回收移植卵巢,对移植体的大小形态、回收率、回收卵母细胞数及其大小进行评价.结果表明:三时间段回收的移植体均生长增大,有腔卵泡明显,回收率达87.5%~92.9%,差异不显著;35 d移植体平均直径达(2 574.7±785.9)μm,显著大于21 d(2 075.3±329.8)μm和28 d(2 088.9±297.7)μm移植体(P<0.01);回收移植体均能分离到卵母细胞,随着移植时间的延长回收卵母细胞数下降,其中21 d移植体回收卵母细胞最多,为(40.5±29.8)个,与35 d移植体回收卵母细胞数(11.1±7.6)个相比差异极显著(P<0.01),28 d移植体回收卵母细胞数(28.4±21.1)个,与前二者差异均不显著;回收卵母细胞形态正常,达到GV期,平均直径达(74.2±2.1)~(75.0±5.9)μm,组间差异不显著;28 d移植体中回收到MⅡ期卵母细胞.随着移植时间的延长,移植体增大,回收卵母细胞数减少.说明在卵巢异位移植中要获得丰富而又充分生长的卵母细胞应根据卵巢卵泡发育规律选择合理的移植时间.
To investigate the influence of transplantation period on the development of oocytes in grafted ovaries in male recipient mice, ovaries from 1-day-old mice were transplanted underneath the kidney capsule of male recipient mice. 21,28 and 36 days after grafted, the ovarian grafts were recovered for morphological evaluation and isolation of oocytes from antral follicles, and the average diameters of grafts and oocytes were determined. The results show the ovarian grafts were successfully recovered with evidence of antral follicle development and increase in size. There was no significant difference in the recovery rates (87.5 % -92.9%) among these three periods (P〉0.05). However, the average diameters of 35 d grafts(2 574.7±785.9) μm was significantly (P〈 0.01) large compared with 21 d (2 075.3±329.8) μm and 28 d grafts(2 088.9±297.7) μm. GV oocytes were retrieved from all grafts recovered and some MII oocytes from 28 d grafts. The number of oocytes recovered decreased with transplantation period extended. Significantly fewer oocytes were retrieved from 35 d grafts (11.1 ±7.6) than 21 d grafts (40.5 ±29.8) (P〈0.01) . There was no significant difference in the average diameters of oocytes from 21-35 d grafts( (74.2±2.1)- (75.0±5.9)μm). In conclusion, transplantation period affects the size of grafts and the number of oocytes produced from ovarian grafts.
