摘要 将含有经过修饰的烟草(Nicotiana tabacumL.)Ⅰ类几丁质酶基因和Ⅰ类β-1,3-葡聚糖酶基因的双价植物表达载体pCG-Ⅱ在农杆菌介导下转化甘蔗优良品种ROC10和ROC22,获得抗G418的转化植株52株,经PCR检测33株呈阳性;对部分经PCR检测呈阳性的转化植株进行Southern杂交和黑穗病抗病性的体外抑菌实验,结果显示这两个基因均已整合到部分甘蔗的基因组中,且它们对甘蔗黑穗病的抗性均有不同程度的提高。 The plant expression vector pCG- Ⅱ containing modified Ⅰchitinase (Chi) gene and Ⅰ 1,3-glucanse (Glu) gene from tobacco was introduced into sugarcane varieties (ROC 10 and ROC22) by Agrobacterium mediated method. After successive G418 resistance selection, 52 transformants were obtained, in which 33 plants were confirmed as transgenic plants by PCR. Some of the PCR-positive plants had been detected by Southern bloting analysis and agar cavity spreading assay for sugarcane smut-inhibition effect. The results indicated that two target genes were integrated into the genomes of some sugarcane plants which exerted different higher inhibitor efficency on the growth of sugarcane smut.
