摘要
目的探讨登革病毒包膜糖蛋白E的结构与功能,为阐明登革病毒感染的分子机制提供科学数据。方法综合利用生物信息学软件(Protean和TMHMM)、数据库(Prosite和PDB)和网络服务器(http://www.expasy.org;http://www.cbs.dtu.dk;http://immune-epitope.org)对E蛋白各级结构及亲水性等多项参数进行分析。结果E蛋白的氨基酸分布情况,二级结构中形成螺旋、折叠、转角、卷曲的氨基酸区段以及包膜糖蛋白E的跨膜区段均得以展示;同时用于预测B细胞表位的柔韧性、表面可及性和亲水性参数得到综合阐释,另外E蛋白的3个结构域和空间结构也在相关数据库中获得。结论生物信息学预测和分析结果可为研究在登革病毒致病及机体免疫应答中E蛋白的结构与功能的关系提供丰富资料。
Objective Glycoprotein E of dengue virus is the major vaccine candidate, Researches on its structure and function are always the highlight, which could elucidate the mechanism of dengue virus infection. Methods Bioinformatics software (Protean and TMHMM ), database (Prosite and PDB ) and net servers (http ://www.expasy.org ; http://www.cbs.dtu.dk; http ://immune-epitope,org)were applied to predict and analyse the structures and biochemical properties of glycoprotein D of dengue virus. Results Amino acid composition and transmembrane helix as well as the secondary structures including regions of helix,sheet,turn,coil of the protein were well defined. Flexibility, hydrophilicity and surface accessibility which are always used to predict the B-cell epitopes were simultaneously displayed, .in addition, three domains and conformational structure of glycoprotein E were obtained from the corresponding database. Conclusion These predicted and analytic results could provide data for understanding the relationship between structure and function of dengue virus in the pathogenesis and the immunological response.
