甘氨酸对肝纤维化及肝硬化大鼠血管活性物质的影响

摘要 目的探讨甘氨酸对肝纤维化及肝硬化大鼠血管活性物质内皮素-1(ET-1),降钙素基因相关肽(CGRP)及心钠素(ANP)的影响。方法将40只雄性Wistar大鼠随机分为5组:正常对照组(C组),肝纤维化模型组(HF组),肝硬化模型组(HC组),肝纤维化甘氨酸干预组(HF+G组)及肝硬化甘氨酸干预组(HC+G组)。观察外周血浆内毒素(endotoxin),丙氨酸氨基转移酶(ALT),血清肿瘤坏死因子-α(TNF-α)水平,采用放射免疫法测定血浆ET-1,CGRP及ANP的量。结果血浆endotoxin、ALT及血清TNF-α水平在HF及HC组明显高于C组(P<0.01),而在HF+G及HC+G组分别低于HF及HC组(P<0.05)。血浆ET-1在HC组高于C组及HF组(分别为P<0.01,P<0.05),在HC+G组低于HC组(P<0.05);CGRP在HF及HC组均高于C组(分别为P<0.01,P<0.05),而在HF+G及HC+G组分别低于HF及HC组(均P<0.05);ANP在HF+G及HC+G组分别高于HF及HC组(均P<0.01)。血浆endotoxin与TNF-α,ET-1,CGRP均呈显著正相关(分别为r=0.523,P<0.01;r=0.467,P<0.01;r=0.417,P<0.05)。结论甘氨酸可减轻肝纤维化及肝硬化大鼠的肝损伤,这可能是通过影响其血管活性物质ET-1、CGRP、ANP的水平实现的。 Objective To explore the effects of glycine on vasoactive substances endothelin-1(ET-1),calcitonin gene related peptide(CGRP)and atrial natriuretic polypeptide(ANP)in rats with hepatic fibrosis and cirrhosis.Methods Forty Wistar rats were randomly divided into five groups:control group,hepatic fibrosis(HF)group,hepatic cirrhosis(HC)group,hepatic fibrosis treated with glycine group(HF+G),and hepatic cirrhosis treated with glycine group(HC+G).The levels of plasma endotoxin,alanine aminotransferase(ALT),serum tumor necrosis factor-α(TNF-α)were detected.The plasma concentrations of ET-1,CGRP,ANP were examined with radioimmunoassay.Results The levels of plasma endotoxin,ALT and serum TNF-α were significantly higher in HF group and HC group than in control group(P〈0.01),but lower in HF+G group and HC+G group than in HF group and HC group,respectively(P〈0.05).The plasma ET-1 was higher in HC group than in control group and HF group(P〈0.01,P〈0.05 respectively),but lower in HC+G group than in HC group(P〈0.05).The CGRP was higher in HF group and HC group than in control group(P〈0.01,P〈0.05 respectively),but lower in HF+G group and HC+G group than in HF group and HC group respectively(P〈0.05).ANP was higher in HF+G group and HC+G group than in HF group and HC group,respectively(P〈0.01).The plasma endotoxin was positively correlated with TNF-α,ET-1 and CGRP(r=0.523,P〈0.01;r=0.467,P〈0.01;r=0.417,P〈0.05,respectively).Conclusion Glycine could lighten liver injuries of rats with hepatic fibrosis and cirrhosis,which may be caused by influencing the levels of vasoactive substances such as ET-1,CGRP and ANP.
