摘要 目的比较单细胞凝胶电泳技术(SCGE)和染色体畸变分析(CA)在遗传毒性检测中的灵敏性。方法48只小鼠分为4组,3个实验组小鼠分别用不同剂量(0.5,1,2mg/kg)的丝裂霉素C(MMC)进行一次性腹腔注射,对照组小鼠注射等体积生理盐水,分别用SCGE和CA对小鼠骨髓细胞的遗传损伤进行检测。结果采用SCGE方法检测,三个MMC剂量组的小鼠骨髓细胞拖尾率与平均尾长均高于对照组,差异有统计学意义;采用CA检测,只有2mg/kgMMC组的畸变细胞率显著高于对照组(P<0.01)。结论对于遗传毒性的检测,SCGE更为敏感,更适用于低剂量致变因子遗传毒性的检测分析。 Objective To compare the sensitivity of single cell gel electrophoresis(SCGE)and chromosome aberrations(CA)in detecting genetic toxicity.Methods Forty-eight mice were divided into 4 groups:three experimental groups and control group.The mice were injected with 0.5,1 and 2 mg/kg Mitomycin-C(MMC)in three groups respectively,and with physiological saline in control group.DNA damage and chromosome aberrations induced by MMC in bone marrow cells of mice were detected by SCGE and CA,respectively.Results The comet frequency and tail length in three MMC groups were significantly higher than in control group(P〈0.05,P〈0.01,P〈0.01),but the frequency of aberration cell in 2 mg/kg MMC group was higher than in control group(P〈0.01).Conclusion SCGE is more sensitive in detecting genetic toxicity.
