摘要
目的:采用基因芯片技术分析不同恶性程度胶质瘤细胞CHG-5(WHO分级为级)和SHG-44(WHO分级为级)的差异表达基因。方法:抽提总RNA,进行RT-PCR,并用荧光染料Cy3和Cy5标记cDNA产物,然后进行芯片杂交,检测CHG-5和SHG-44基因表达的差异,并用Northern blot杂交来验证芯片结果。结果:与CHG-5相比,SHG-44细胞中检测到有103个基因表达明显上调,89个表达明显下调。芯片结果得到Northern blot杂交结果的支持。结论:初步揭示了胶质瘤进展的差异表达基因,为胶质瘤的进一步研究提供了基础资料。
Objective:To identify the differentially expressed genes in glioma cell lines CHG-5 (WHO grade Ⅱ ) and SHG-44 (WHO grade IV ) using cDNA microarray. Methods :Differentially expressed genes of glioma cell lines SHG- 44 and CHG-5 were detected by cDNA microarray. Total RNA of the cell lines were extracted and reverse-transcripted. The cDNA product was labeled with Cy3&Cy5 fluorescence dye for microarray hybridizations. NOrthern blot hybridizations were used to confirm the differentially expressed genes detected by the microarrays. Results :Compared with the cell line CHG-5, 103 genes were up-regulated (ratio:〉2.0) and 89 genes were down-regulated (ratio〈0.5) in the SHG-44 cell line. Conclusion: The differentially expressed genes may be involved in the progression of glioma, they can provide the basic data for further research the neurogliocytoma.
