高糖、高脂肪酸饮食对大鼠骨骼肌葡萄糖转运体4 mRNA表达的影响 被引量:1

摘要 成年Wistar大鼠随机分为对照组、高糖组、高饱和脂肪酸组、高不饱和脂肪酸组,喂养24周后,各实验组与对照组相比葡萄糖输注率(GIR)明显减低(P<0·01),大鼠骨骼肌葡萄糖转运体4(GluT4)mRNA的表达显著降低(P<0·01),其表达与游离脂肪酸呈明显负相关,与GIR呈明显正相关。高糖、高饱和脂肪酸及高不饱和脂肪酸饮食均可造成大鼠骨骼肌GluT4 mRNA的表达降低,这可能是胰岛素抵抗的原因。 To explore the effect of high sucrose(HS),high saturated fatty acid(HSFA) and high unsaturated fatty acid(HUFA) diets on insulin resistance (IR) and the expression of GluT4 protein in skeletal muscle. Adult Wistar rats were devided into groups of normal control (NC), HS , HSFA , HUFA. And IR was evaluated by glucose infusion rate (GIR) of clamp technique after 24 weeks of diet treatment. The mRNA expression of GluT4 was measured by RT-PCR. GIR was obviously lower in experimental groups than that in NC group. The expression of GluT4 mRNA in skeletal muscle was markedly decreased in all experimental groups compared with NC. The expression of GluT4 in skeletal muscle was negatively correlated with free fatty acid (FFA) and well positively correlated with GIR. The expression of GluT4 mRNA in skeletal muscle is decreased in high sucrose and high fat diets rats, and obviously correlated with plasma FFA negatively and with GIR positively.
