摘要
目的观察丁酸钠对大鼠小肠上皮细胞株~6(intestinal epithelium cell line No.6,IEC-6)细胞增殖和凋亡的作用。方法不同浓度丁酸钠(低浓度2mmol/L,中浓度8mmol/L,高浓度14、20mmol/L)处理IEC-6细胞,细胞计数试剂盒CCK-8法检测细胞增殖情况,分光光度法检测细胞内Caspase-3活性程度,流式细胞术检测细胞凋亡率。结果(1)加入丁酸钠(2、8、14、20mmol/L)共培养24h后,2、8mmol/L丁酸钠组细胞增殖速度比对照组快,差异有统计学意义(P〈0.01);48h后,14、20mmol/L丁酸钠组细胞增殖较对照组明显受抑制,差异有统计学意义(P〈0.01);72h后,8、14、20mmol/L丁酸钠组增殖亦明显受抑制,与对照组比较差异有统计学意义(P〈0.01)。(2)加入丁酸钠(2、8、14mmol/L)共培养12h后,8、14mmol/L丁酸钠组与对照组比较Caspase-3活性明显升高,差异有统计学意义(P〈0.05);24h后,2、8、14mmol/L丁酸钠组与对照组比较Caspase-3活性均升高,差异有统计学意义(P〈0.05);48h后,8、14mmol/L丁酸钠组与对照组比较Caspase-3活性亦升高,差异有统计学意义(P〈0.05)。(3)加入丁酸钠(2、8、14mmol/L)共培养12、24、48h后,8、14mmol/L丁酸钠组与对照组比较IEC-6细胞凋亡率均有升高,差异有统计学意义(P〈0.05)。结论中、高浓度丁酸钠有诱导IEC-6细胞凋亡,抑制IEC-6细胞增殖作用,而低浓度丁酸钠没有这种作用。
Objective To investigate the effects of sodium butyrate on the proliferation and apoptosis of intestinal epithelium cell line No. 6 (IEC-6)of the rats. Methods IEC-6 cells were treated with sodium butyrate at different concentrations (low concentration 2 mmol/L, medium 8 mmol/L, high 14 and 20 mmol/L) or incubated in medium alone as a control. Morphological changes of IEC-6 cells were observed by inverted microscope, and cell proliferation was assessed by cell counting kit-8 (CCK-8) assay. Caspase-3 activity was measured and flow cytometry was used to assay the apoptosis. Results (1) The growth rates of the ceils treated with sodium butyrate in concentrations of 2, 8 mmol/L for 24 h were higher than those of control group (P〈0. 01) . At 48 h, the 14, 20 mmol/L sodium butyrate groups had the lower growth rates than control group (P〈 0.01). At 72 h, the 8,14, 20 mmol/L sodium butyrate groups had the lower growth rates than control group (P〈0.01). (2) The caspase-3 activity from cell lysates of IEC-6 cells treated with sodium butyrate in concentrations of 8, 14 mmol/L for 12 h were higher than that of control group (P 〈0.05) . At 24 h, the 2, 8,14 mmol/L sodium butyrate groups had the higher caspase-3 activity than control group (P〈0.05) ; At 48 h, the 8, 14 mmol/L sodium butyrate groups had the higher caspase- 3 activity than control group (P〈0. 05). (3) The rates of cell apoptosis in groups treated with sodium butyrate in concentrations of 8, 14 mmol/L for 12, 24, 48 h were higher than those of control group (P〈0.05). Conclusions Sodium butyrate induces apoptosis and inhibits the proliferation in rat intestinal epithelial cells, and the effects are dose and time dependent.
