以铽-诺氟沙星络合物作为荧光探针测定胆汁酸的研究

摘要 以Tb^3+-诺氟沙星络合物作为荧光探针,提出了一种新的荧光光度法测量胆汁酸(BA)的方法。在pH=9.00的酸度条件下,BA能使铽(Tb^3+)-诺氟沙星(NF)二元络合物中Tb^3+位于545nm处的荧光强度增强达18倍,且增强的荧光强度与胆汁酸的含量成正比。测量的线性范围为5.00×10^-7 ~2.00×10^-6 mol L^-1,检测限为2.40×10^-8mol L^-1,回收率为93.3%~103.7%,RSD为0.60%。该方法成功地应用于血清实际样品的测定。 Bile acid (BA) in serum was determined using the luminescent terbium norfloxacin bile acid (Tb^3+- NF- BA) complex. The characteristic fluorescence of Tb^3+ at 545 nm in Tb^3+-NF binary complex is observed and the fluorescence intensity increases up to 18-fold if BA is added to the binary complex. The enhanced fluorescence intensity is in proportion to the concentration of BA. The linear range of the developed method is from 5.00 × 10-7 to 2.00 × 10^-6 mol L-I and detection limit is 2.40 × 10^-8 mol L-I of BA using Tb^3+-NF as fluorescent probe. Some Organic compounds and albumin in serum samples were removed in order to improve the selectivity of the proposed method. The recovery of this method was over the range of 93.3 to 103.7% and the relative standard deviation was 0.60%. The developed method was successfully applied to the determination of BA in serum samples.
