摘要
目的建立一种新的血清单胺氧化酶(MAO)测定法,以提高检测MAO活力的特异性和准确性。方法用3-甲基-N-乙基(β-羟乙基)苯胺(MEHA)为色原系统进行患者血清MAO活力测定。结果对缓冲液最适浓度、pH和种类的选择非常重要,可提高灵敏度,线性范围为0~250U/L,正常值范围为30U/L内,回收率为92.4%~102.6%,精密度:批内CV=2.5%~4.0%,批间CV:3.3%~4.5%。与二硝基苯肼比色法比较,相关性良好(r=0.986)。结论本法具有高度的特异性和准确性,操作简便,灵敏快速,标本用量少,结果稳定,对肝硬化的诊断有非常重要的作用。
Objective To develop a novel assay for serum monoamine oxidase (MAO) determination, so as to improve the specificity and accuracy of MAO activity measurement. Methods Serum MAO activity was measured with 3-methy N-ethyl (β-hydroxyethyl) aniline (MEHA) as pigment system. Results It was vital to select optimal concentration, pH value and sorts of buffers. The established method improved the detecting sensitivity. The corresponding parameters were as follows: the linear range:0-250 U/I.; the reference range: 〈30 U/L; the recovery rate: 92.4%-102.6%; exactitude: within-run CV= 2.5%-4.0% ; between-run CV=3.3%-4.5%. The method was closely correlated with dinitro-phenylhydrazine chromatometry (r=0. 986). Conclusion The novel method has high specialty and accuracy, and it is characteristic of convenience, sensitivity, rapidity, less sample and re sult stability, contributes to the diagnosis of liver cirrhosis.
