摘要
目的:运用Fura-2/AM荧光法测定S-180肉瘤细胞内钙离子浓度的方法,观察猪苓多糖(polyporusum bellatus,PB)、茯苓多糖(pachyman polysaccharide,PPS)和黄芪多糖(astragalus polysaccharide,APS)对细胞内Ca2+浓度的影响。方法:BALB/c小鼠腹水扩增获得的S-180肉瘤细胞中分别加入不同质量浓度的3种中药多糖分别为100、250和500μg/mL,对照组加入相应体积的灭菌的0.9%的氯化钠溶液。培养1和8h后,采用Fura-2/AM荧光法测定培养S-180肉瘤细胞中的Ca2+浓度。结果:PB和PPS在作用1和8h后能使S-180肉瘤细胞内的Ca2+浓度升高,与对照组相比差异有统计学意义(P<0.05);PPS高剂量组与中低剂量组相比差异有统计学意义(P<0.05);APS在作用1和8h后S-180肉瘤细胞内Ca2+浓度的变化与对照组相比差异无统计学意义(P>0.05)。结论:PB和PPS能引起S-180肉瘤细胞内Ca2+浓度的升高,而APS则不能引起S-180肉瘤细胞内Ca2+浓度的升高。
Objective: To study the effects of polyporusum bellatus ( PB), pachyman polysaccharide (PPS) and astragalus polysaccharide (AlaS) on cytoplasmic free calcium in S-180 cells. Methods: S-180 cells were obtained from the malignant ascites of BALB/c mice. The S-180 cells were incubated with the three polysaccharides at 100,250 and 500 μg/mL for 1 and 8 h. The concentration of intracellular free calcium in S-180 cells was measured by Fura-2/AM. Results: The intracellular calcium concentration increased in S-180 cells at 1 and 8 h after incubation with PB and PPS. The difference was significant compared with control (P 〈0.05 ). There was significant difference between PPS high dosage group and PPS low and median dosage groups ( P 〈 0.05 ). But treatment with Alas for 1 and 8 h had no significant effects on intracellular calcium concentration compared with control (P 〉 0.05 ). Conclusion: PB and PPS increases the intracellular calcium concentrations in S-180 cells. Alas does not induce the increase in intracellular calcium concentratlon.
