人黑色素瘤相关抗原基因-12基因疫苗对Lewis肺癌小鼠肿瘤生长及体内抗体水平的影响 被引量:2

摘要 目的:研究人黑色素瘤相关抗原基因-12(MAGE-12)基因疫苗pEGFP-C3-MAGE-12对Lewis肺癌小鼠肿瘤生长及体内抗体水平的影响。方法:小鼠80只,随机分为4组,每组20只。疫苗组、空质粒组和生理盐水(NS)组接种Lewis肺癌瘤组织制备Lewis肺癌小鼠模型。接种癌组织前14d、7d、0d,疫苗组按每次每只0.2mL(75ng/L)于右下肢内侧肌肉群内注射pEGFP-C3-MAGE-12;空质粒组和NS组每只每次注入pEGFP-C3空质粒或NS;空白对照组小鼠不做任何处理。每3d观察并记录1次肿瘤大小。接种瘤组织后14d每组取10只小鼠,ELISA法检测血清MAGE-12-Ab,余小鼠(每组10只)自然死亡,记录生存时间。结果:3组7d成瘤率均为100%。空质粒组、NS组及疫苗组生存期分别为(27.0±0.3)d,(27.2±0.3)d和(28.9±0.6)d;肿瘤体积分别为(179.775±9.085)mm3,(173.172±9.085)mm3和(144.923±9.528)mm3;疫苗组血清MAGE-12-Ab水平为(711.16±174.78),其他3组未检测到MAGE-12-Ab;与空质粒组、NS组比较,疫苗组生存期短(P<0.05),肿瘤体积小(P<0.05),体内抗体水平高(P<0.05)。结论:MAGE-12基因疫苗具有免疫治疗作用。 Aim: To study the role of treating mouse lung cancer model with immunotherapy of melanoma associated antigen gene-12 (MAGE-12) DNA vaccine. Methods:A total of 80 mice were equally randomized into 4 groups. The vaccine group, pEGFP-C3 group and NS group were injected tumor tissue suspension of Lewis lung cancer into the right medial muscle of lower limb to establish the animal model. 14 days,7 days, and 0 day before tumor tissue suspension injection, groups mentioned above were injected pEGFP-C3-MAGE-12, pEGFP-C3 and normal saline at the same muscles,respectively. The blank control group were injected nothing. The growth of tumor and serum MAGE-12 antibody in mice were investigated. Results:In pEGFP-C3 group, NS group and vaccine group, the survival time were (27.0 ±0.3 ) d, (27.2±0.3 ) d, and ( 28.9 ± 0.6 ) d ; the size of tumor were ( 179. 775± 9. 085 ) mm^3 , ( 173. 172 ± 9. 085 ) mm^3 , and ( 144. 923 ± 9. 528 )mm^3 ; the level of MAGE-12 antibody of vaccine group was( 711.16 ± 174.78) ,while MAGE-12 antibody of the other groups were not detected. Compared with those of pEGFP-C3 group, NS group and the blank control group, the survival time of vaccine group was longer, the size of tumor was smaller(P 〈 0.05) , the serum level of MAGE-12 antibody was higher( P 〈 0.05). Conclusion: MAGE-12 gene vaccine can treat lung cancer by immunotherapy.
