葡萄籽中原花青素灌胃及注射给药的抗炎作用和刺激性 被引量:1

Anti-inflammatory effects of procyanidin from grape seeds by intragastric and injective administrations

摘要 目的研究葡萄籽中原花青素(proanthocyanidins,PA)灌胃给药的抗炎作用及注射给药的刺激性。方法用巴豆油诱导的小鼠耳肿和角叉菜胶诱导的大鼠足肿模型,观察原花青素灌胃给药的抗炎作用,并观察原花青素肌肉和腹腔注射的刺激性。结果PA 40 mg/kg单次腹腔注射能明显抑制角叉菜胶诱导的大鼠足肿和巴豆油诱导的小鼠耳肿,而PA 200~400 mg/kg单次或多次灌胃给药对角叉菜胶诱导的大鼠足肿和巴豆油诱导的小鼠耳肿均无明显影响,但给药期间小鼠和大鼠均未见明显异常反应;PA 40 mg/kg腹腔注射后小鼠活动减少、蜷缩,部分小鼠出现扭体反应,肌肉注射24h后给药部位有渗出液、充血、组织坏死等反应。说明PA腹腔和肌肉注射有刺激性。结论葡萄籽中原花青素灌胃给药无明显的抗炎作用,而注射给药有一定的刺激性。 Objective To study the anti-inflammatory effects of procyanidin (PA) from grape seeds by intragastric and injective administrations. Methods Croton oil-induced ear swelling model of mice and carrageenan-induced hind paw edema model of rats were established, and the anti-inflammatory effects of PA were investigated by intragastric while its injective irritation was investigated by muscular and intraperitoneal injections. Results PA 40 mg/kg peritoneal injection once significantly inhibited carrageenan-induced paw edema in rats and croton oil-induced ear swelling in mice, but PA 200-400 mg/kg intragastric once or 7 times successive administration had no anti-inflammatory effects and paradoxical reactions. After 40 mg/kg peritoneal injection, some mice appeared underactivity, scrunch, and writhing. The administration sites appeared congestion, diffusate and necrosis when administered by 40 mg/kg intramuscular injection after 24 h. Conclusion PA has no anti-inflammatory effects on experimental inflammation in rats and mice by intragastric administration, but irritation exists after intramuscular injection or peritoneal injection.
