交变磁场介导下半乳糖白蛋白磁性阿霉素对人肝癌细胞和肝细胞的影响

Effects of galactosed human serum albumin magneticadriamycin nanoparticles on human hepatoma cell HepG2 and human liver cell L-02 in vitro combined with an alternative magnetic field

摘要 目的观察半乳糖白蛋白磁性阿霉素纳米粒联合交变磁场对肝癌细胞和人肝细胞的影响。方法将人肝癌细胞HepG2及人肝细胞L-02细胞,随机分成A1、A2组(即RPMI 1640对照组),B1、B2组(游离阿霉素),C1、C2组(半乳糖白蛋白磁性阿霉素纳米粒),D1、D2组(阿霉素纳米粒),E1、E2组(半乳糖白蛋白磁性阿霉素纳米粒),实验组每组含有0.5mg/L或等量的阿霉素;除C1、C2组外,其他各组置交变磁场(频率为100kHz、磁场强度为15kA/m)中,作用30min。通过细胞计数、Hoeehst荧光染色、DNA蛋白电泳、流式细胞仪检测,观察细胞生长曲线、细胞形态、细胞凋亡及细胞周期的变化。结果(1)E1、E2组,HepG2和L-02细胞增殖抑制作用最强,DNA梯形带最明显,细胞凋亡率分别达40.12%、45.3%;S期细胞分别为39.53%、53.46%,明显高于其他组(P〈0.01);(2)L-02细胞的形态改变、细胞增殖抑制、细胞凋亡率明显强于HepG2细胞(P〈0.05)。结论半乳糖白蛋白磁性阿霉素纳米粒在交变磁场介导下,对HepG2和L-02细胞产生热化疗协同增效的作用。 Objective To observe the effects of Gal-MADM on human hepatoma cell HepG2 and human liver cell L-02 combined with alternative magnetic field. Methods HepG2 cells and L-02 cells were randomly divided into 5 groups, respectively: A1, A2 ( control group ), B1, B2 (adriamycin) ; C1, C2 ( galactosed human serum albumin magnetic adriamycin nanoparticles, Gal-MADM) ; D1, D2 ( adriamycin nanoparticles, ADM-NP) ; El, E2 ( Gal-MADM-NP). Each of experimental groups was administered with 0. 5 mg/L or equal adriamycin. Except group C1 and C2, the other groups were exposed to an alternative magnetic field ( AMF ) with 100 kHz, 15 kA/m for 30 min, respectively, The growth curve of cell, cyctomorphologic changes, DNA fragmentation, cell cycle and apoptosis were assayed by cell count,fluorescence microscope ,agarose gel elctrophoresis and flow eytometry. Results (1) the rate of proliferation inhibitation was highest,the typical DNA ladder was most obvious, the apoptosis ratio of hepG2 cell and L-02 cell was 40.12% and 45.32% ,the percentage of both cells in phase S was 39.53% ,53.46% ,respectively in the group E1, E2 and obviously higher than the other groups (P 〈 0.01 ). (2) apoptosis ratio, the percentage of cells in phase S of L-02 cells were higher ( P 〈 0.05) and alternation of the apoptotic cytomorphology more obvious than HepG2 cells. Conclusion With the mediation of AMF, Gal-MADM exert a coordinative effect on HepG2 cells and L-02 cells and L-02 cells.
