摘要 目的观察脂多糖(LPS)体外作用于大鼠肝星状细胞(HSC)时肿瘤坏死因子-α(TNF-α)和结缔组织生长因子(CTGF)表达变化,探讨TNF-α对LPS诱导的HSC中CTGF表达的影响。方法用一步密度梯度离心法分离大鼠HSC,体外培养LPS处理HSC,逆转录.聚合酶链反应(RT-PCR)方法检测不同LPS浓度处理的HSC CTGF mRNA表达水平,并用ELISA双抗体夹心法测定TNF-α浓度。结果TNF-α水平随LPS浓度增加而增加,LPS可上调CTGF mRNA的表达,TNF-α浓度及CTGF mRNA表达水平呈一定的平行关系。结论LPS可上调HSC中CTGF mRNA表达,该过程可能是通过TNF-α介导的。 Objective To explore the effect of lipopolysaccharide (LPS) on the expression of connective tissue growth factor (CTGF) in rat hepatic stellate cells (HSC) in vitro and the effect of tumor necrosis factor-α (TNF-α) on the expression of CTGF in HSC induced by LPS. Methods Rat HSCs were separated by in situ perfusion of pronase and collengase, and single-step Nycedenz density gradient centrifugation. After in vitro culture and passages, HSCs were treated by E coli-derived LPS. The CTGF mRNA expression was detected by simiquantitative RT-PCR with different LPS concentrations. TNF-α was determined by enzyme-linked immunosorbnent assay. Results The level of TNF-α was increased with the increased concentrations of LPS, and LPS could also up-regulate the expression of CTGF mRNA and there existed a parallel relationship. Conclusion LPS can up-regulate the expression of CTGF mRNA of HSCs, and TNF-α might play an important role in the pathological process of the expression of CTGF mRNA.
