Establishment of nicotine experimental periodontitis model in rats

摘要 目的:建立大鼠实验性牙周炎动物模型并观察尼古丁对其的影响。方法:采用36只SD大鼠,丝线结扎上颌右侧第二磨牙颈部,左侧未结扎作为自身对照。随机分为生理盐水对照组、尼古丁每日低剂量组和高剂量实验组,分别给予生理盐水、尼古丁每日0.83mg/kg、1.67mg/kg腹腔注射。给药第14、28天处死动物,取材。采用组织学、Micro-CT方法观察大鼠牙周破坏情况。结果:第14、28天各组大鼠结扎侧第二磨牙龈沟出血指数(SBI)、牙周探诊深度(PD)、牙槽骨丧失值(ABL)均高于对照侧(P<0.01),尼古丁给药后各测量值进一步加重,并呈浓度依赖性。仅给予尼古丁亦可造成大鼠非结扎侧牙槽骨吸收。结论:组织学、Micro-CT方法证实牙颈部丝线结扎方法可成功的建立牙周炎实验模型,腹腔注射尼古丁可致加重大鼠牙槽骨丧失。 AIM: To evaluate the effect of nicotine administration on periodontal breakdown resulting from ligature - induced periodontitis in rats. METHODS : Thirty - six adult male SD rats were received silk ligatures around the cervixs of the right second maxillary molar while the contralateral tooth was left unligated. Then the animials were assigned randomly to daily subcutaneous injections of saline solution ( control, group A) or nicotine ( group B and C). The animals were sacrificed at days 14 and 28 after ligature placement. Histological and microcomputed tomography (Micro -CT) examinations were used to evaluate the periodontal breakdown. RESULTS: Ligation significantly increased sulcular bleeding index (SBI) , periodontal depth (PD) and alveolar bone loss (ABL) of the right second maxillary molars ( P 〈 0.01 ). Nicotine administration enhanced the alveolar bone loss in a dose - dependent way. CONCLUSION: Histological and Micro - CT measurements confirmed that ligature can successfully induce experimental periodontitis in rats. Daily administration of nicotine resuhes in significantly greater alveolar bone loss.
