新型维A酸AHPN促人皮肤恶性黑素瘤A375细胞凋亡及机制研究

摘要 目的:探讨新合成维A酸AHPN对体外培养的皮肤恶性黑素瘤细胞系A375的增殖抑制及诱导凋亡作用。方法:噻唑蓝(MTT)法测定AHPN对A375细胞系的增殖抑制作用,光镜观察药物对细胞形态的影响,用流式细胞术研究药物对细胞凋亡及细胞周期的影响。基因系统测定体外A375细胞中AHPN作用与核转录因子NF-κB活化关系。结果:维A酸AHPN可有效抑制A375细胞的生长,并具有剂量依赖性。AHPN可促进A375细胞的凋亡及G1期抑制;可增强A375细胞NF-κB活性。结论:AHPN可有效抑制A375细胞的增殖并诱导其凋亡。AHPN对A375细胞的促凋亡作用与NF-κB活化有关。 Objective: To investigate the effects of AHPN on the apoptosis and anti- proliferation in human skin malignant melanoma cell A375 in vitro. Methods: Thiazolyl blue (MTF) assay was used to determine the anti - proliferation of AHPN on A375 cells. Flow cytometry was performed to determine the effect of AHPN on cell cycle and cell apoptosis. Transfection and luciferase assays were performed to associate nuclear factor- κB (NF- κB) with pro - apoptotic function of AHPN. Results: AHPN effectively inhibited the proliferation of A375 cells in a concentration - dependent manner, promoted marked apoptosis and G1 arrest in A375 cells and increased the activity of NF - κB. Conclusion: AHPN can effectively inhibit proliferation and induce apoptosis in A375 cells. The pro-apoptotic function of AHPN is related to NF- κB activation.
