摘要
目的 研究TGF-β1基因多态性在SARS-冠状病毒(SARS-CoV)感染及愈后肺间质纤维化中的作用。方法 选取65例SARS康复人员、37例医护人员和66例健康人员,进行TGF-β1基因多态性与SARS-CoV易感性及与SARS愈后肺间质纤维化关系的分析。采用病例一病例研究设计,在SARS康复患者中分析TGF-β1基因多态性与SARS愈后肺间质纤维化的关系。采用聚合酶链反应一直接测序法(PCR-SBT)进行TGF-β1基因多态性检测,数据行t检验和卡方检验。结果 TGF-β1基因+869和+915位点的基因多态性在SARS康复人群、医护人员和对照人群分布差异无统计学意义,未见该基因两位点的多态性连锁与SARS-CoV易感性有关。SARS愈后人群的肺间质纤维化改变与TGF-β1基因+869和+915位点的基因多态性无关,也未见该基因+869和+915位点的多态性连锁与肺间质纤维化相关。结论 TGF-β1基因+869和+915位点的基因多态性可能与SARS—CoV易感性无关;SARS康复人员的肺间质纤维化可能与TGF-β1+869和+915基因多态性无关。
Objective To investigate the roles of transforming growth factor(TGF)-β1 gene polymorphisms in severe acute respiratory syndrome-coronavirus (SARS-CoV) infection and the interstitial lung fibrosis after recovered. Methods Sixty-five recovered SARS patients, 37 health care workers and 66 healthy controls were enrolled in this case-case study. The association between genetic polymorphisms of TGF-β1 and susceptibility to SARS or interstitial lung changes after SARS recovered was carried out. Polymerase chain reaction-sequencing based typing(PCR-SBT) method was used to determine the polymorphisms of TGF-β1 gene at locus +869 and +915. Data were analyzed using t test and chi square test. Results There was no significant association of TGF-β1 gene polymorphisms at locus + 869 and + 915 in recovered SARS patients, health care workers and healthy controls. And gene linkage of this two loci was not related with SARS-CoV susceptibility. Furthermore, no association between interstitial lung changes in recovered SARS patients and TGF-β1 gene polymorphisms or genetic linkage of this two loci. Conclusions It may not be related between TGF- β1 gene polymorphisms at locus +869 and +915 and SARS-CoV susceptibility. And interstitial lung changes in recovered SARS patients may not be influenced by TGF-β1 gene polymorphisms.
