何首乌提取物二苯乙烯苷对PQ-MB诱导小鼠黑质多巴胺能神经元的保护作用 被引量:4

Neuroprotective effects of TSG on Degeneration of Nigral Dopaminergic Neurons Induced by Paraquat and Maneb in Mice

摘要 目的:检测何首乌提取物二苯乙烯苷(2,3,5,4-tetrahydroxystilbene-2-O-β-D-glu-coside,TSG)对百草枯(Paraquat,PQ)和代森锰(Maneb,MB)诱导的小鼠黑质多巴胺能神经元凋亡的保护作用及机制。方法:C57BL/6小鼠腹腔注射PQ及MB制备帕金森病模型,治疗组小鼠口服TSG,分别为大(160mg/kg)、中(80mg/kg)、小(40mg/kg)3个剂量组。六周后用酪氨酸羟化酶(TH)免疫组织化学法、尼氏染色检测多巴胺能神经元的凋亡,免疫组织化学法检测黑质多巴胺能神经元Caspase-3活化情况。结果:TSG大、中剂量组的尼氏阳性神经元及TH阳性神经元脱失明显减少,Caspase-3活化率明显降低。结论:一定剂量的TSG对PQ和MB联合诱导的小鼠黑质多巴胺能神经元凋亡有保护作用。 Objective: To observe the role of TSG (2, 3, 5, 4 - tetrahydnystilbene - 2 - O - β - D - glucoside) in preventing against PQ (paraquat) and MB (maneb) -induced apoptosis of nigral dopaminergic neurons in mouse model and the related mechanisms. Method: C57BL/6 mice were intraperitoneal injected PQ and MB to erect animal model of Parkinson's disease. Mice in 3 TSG protective groups (high, middle, low concentration group) were adiministrated with different doses of TSG (40mg/kg, 80mg/kg, 160mg/kg) , respectively. 6 weeks later, Nisal staining, tyrosine hydroxylase (TH) immunostaining were used to observe the apoptosis of nigral neurons and immunohistochemical staining was carried to detect Caspase - 3 activity, Results: In TSG groups of high and middie concentrations, the loss of nissl positive and TH positive - neurons were decreased significantly, and Caspase - 3 activity was also reduced obviously. Conclusion: Certain concentrations of TSG showed protective effects on apoptosis of nigral dopaminergic neurons induced by PQ and MB in mice.
