摘要
目的8-异前列腺素F2a(8-iso—PGF2a)作为反映缺氧缺血-再灌注后脂质过氧化作用的指标具有准确、特异、敏感的特性。而新型抗氧化剂N-乙酰半胱氨酸(NAC)具有清除氧自由基抗氧化的功能。本研究旨在探讨实验性缺氧缺血性脑损伤大鼠尿与脑组织8-iso—PGF2a相关性及尿8-iso—PGF2a能否作为监测早期脂质过氧化作用的生化指标及NAC的治疗效果。方法7日龄SD新生大鼠60只随机分为对照组、缺氧缺血组和NAC组。NAC组给予NAC0.1mg/(g·d),于缺氧缺血前3d灌胃,共3d;缺氧缺血组给予等体积生理盐水,对照组不做相应处理。应用ELISA法测定各组大鼠尿与脑组织8-iso—PGF2a含量。结果缺氧缺血组尿和脑组织含量分别为(101.5±22.8)ng/mmol.cr和(237.2±11.3)ng/g,明显高于对照组(26.5±11.6)ng/mmol.cr和(90.8±18.7)ng/g(P〈0.01),NAC组(40.1±9.9)ng/mmol.cr和(110.5±12.4)ng/g,明显低于缺血组(P〈0.01),尿与脑组织8-iso—PGF2a水平相关(r=0.823,P〈0.01)。结论急性缺氧缺血性脑损伤时尿8-iso—PGF2a含量产生增加,NAC能有效的降低尿8-iso—PGF2a水平,可用于治疗缺氧缺血性脑损伤。
Objective 8 -iso- prostaglandin F2a (8 -iso- PGF2a) is an index that can sensitively,truthly and specificany reflect peroxidation caused by reperfusion. N- acetlycysteine(NAC) acts as an effective scavenger for free radical cleaning in the body. This study aimed to analyze the correlation of 8 - iso - PGF2a level between urine and brain tissue, and to investigate the rapeutic effect of NAC, and the possibility of level of 8 - iso - PGF2a in urine as biochemicalindex. Methods Seven - day - old Spraygue - Dawley(SD) rats were randomly divided into 3 groups : hypoxia - ischemia, NAC and control group. Rats in the NAC group were given dastric lavage with NAC 0. 1 mg/( g · d) for three days, Rats in hypoxia- ischemia group were given an equal volune of normal saline, and control group had no administra -tion. The 8 -iso -PGF2a contents in urine and brain tissue were determined by ELISA. Results Compared with the control group, 8 - iso - PGF2a contents in urine and braintissue of the hypoxia - ischemia group were significantly higher [ (26. 5 ± 11. 6 ) ng/mmol, cr vs ( 101.5 ± 22.8 ) ng/mmol, cr and ( 90. 8 ± 18.7) ng/g vs(237. 2 ± 11.3) ng/g] ,both P 〈0. 01. In the NAC group, 8 - iso - PGF2a concentration of urine and brain tissue were (40.1 ± 9. 9 ) ng/mmol, cr and ( 110. 5 ± 12.4 )ng/g respectively, lower than those in the hypoxia - ischemia group( P 〈 0. 01 ). The 8 - iso - PGF2a levels of urine and brain tissue were positively correlated ( r = 0. 823, P 〈 0. 01 ). Conclusions Thelevel of 8 - iso - PGF2a increased in both urine and brain tissue of rats with acute brain hypoxia - ischemia, NAC reduces the level of 8 - iso - PGF2a in urine. Therefore it may be used in treatment of brain hypoxia - ischemia.
