选择性酸降解叶绿素产物分析及与牛血清白蛋白的相互作用

摘要 脱镁叶绿酸a可以作为光敏活性剂用于光动力治疗.从天然植物的叶绿素出发,采用酸选择性降解,硅胶层析纯化的方法制备脱镁叶绿酸a.得到的产物经液相色谱、二极管阵列检测和质谱分析,表明其光谱吸收和分子量均与脱镁叶绿酸a一致.利用荧光光度法研究了脱镁叶绿酸a与牛血清白蛋白的相互作用,结果表明:脱镁叶绿酸a与牛血清白蛋白形成1:1的配合物,结合常数为5·0×105L/moL. As a photosensitizer pheophorbide a (Pheide a) is used in photodynamic therapy. In this study, preparation of pheide a from a chlorophyll mixture utilizing the selective hydrolysis and silica chromatography separation was investigated. The analysis of product using liquid chromatography-mass spectrometry ( LC - MS) with photodiode array detector showed that its absorption spectra and molecular weight were the same with pheide a. Its interaction with bovine serum albumin(BSA) was studied with fluorescence spectra. The results indicated that pheide a bound to BSA with a 1:1 adduct formed ,which caused the static fluorescence quenching of BSA. The binding constant is 5.0 × 10^5 L/mol.
