摘要
海栖热袍菌(Thermotoga maritima MS8)是一种极端嗜热厌氧菌,细胞始终被一种鞘结构所包围,并且鞘在细胞两端呈现气囊状.通过膜截留法、透析法和等密度梯度离心法对鞘进行提纯,其中密度梯度离心法提纯的效果较理想.通过TLC分析,鞘由糖、蛋白、脂类组成,偏向膜的性质.通过对鞘的敏感性研究,发现蛋白酶K可以帮助鞘形成孔洞,并且仍可保留细胞的完整性,这可能有助于进一步促进T.maritima MS8基因转化工作的发展.
Thermotoga maritima MS8 is strictly anaerobic and extremely thermophilic. The rod-shaped cells are surrounded with sheath like structure ballooning over the ends of the rods. In contrast to the application of centrifugational membrane filter or dialysis, the method of sucrose isopycnic gradient centrifugation was more suitable. Through the analysis of thin-layer chromatography, sheath was composed of proteins, lipids and saccharide. It resembled the characterization of outer membrane. And sheath was sensitive to Proteinase K while the cell was still alive. This finding may promote the transformation study of Thermotoga maritima.
