摘要
原生质体瞬间表达系统一般用于研究特定基因5′和3′端不同序列在相应基因表达中的作用、不同启动子对于特定基因的转录活性及为外源基因最终导入植株摸索实验条件,而将其用于研究动物基因在植物细胞中的表达情况则少见报道。
Human defensin-1 (human neutrophil peptide-1, HNP-1) is one of the small peptides found recently in human neutro-phils with highly antimicrobial activity of broad-spectrum. It is greatly significant to express cDNA of this peptide in higher plants to produce novel antipatho-genetic crops. In this work, a group of vector with the promoter of different activity, with or without some part of prepropiece were constructed and transfe-red to tobacco mesophyll protoplast. The result showed that HNP-1 expression vectors with some part of propiece (from
Hind Ⅲ) could transiently express HNP-1 in tobacco protoplast; the expression level of those with the promoter of high activity was higher than that of those with the promoter of low activity and the expression peak of the former was faster to be got. The expression vector with total prepropiece could not transiently express HNP-1 in tobacco protoplast. According to this result, it was good for the expression of HNP-1 in plants to choose for a more active promoter and delete the pre and some part of propiece from HNP-1 cDNA.
