摘要
以普通红豆草的下胚轴、愈伤组织为材料,研究了不同的酶种类及组合、酶液浓度、酶解时间等因子对其原生质体游离的影响。结果表明,愈伤组织作为原生质体游离材料好于下胚轴;红豆草愈伤组织在1.0%cellulose+0.8%Macerase+0.5%macerozyme R-10的酶液中酶解6 h为较理想的原生质体酶解条件。
Using hypocotyl and its induced callus as tested materials, the various influencing factors on protoplast dissociation were studied,such as the kinds of enzyme,the concentration and combination of enzyme,dissociation-time. The results showed the callus was better than hypocotyl as material of protoplast dissociation;it was in better condition of protoplast dissociation that the callus were treated in a mixture of 1.0% cellulose+0.8% Macerase+0.5% macerozyme R-10 for 6 h.
