摘要
乏氧是许多疾病的病理生理改变,乏氧促进了肿瘤的恶性表型和侵袭性行为及转移的发生,导致肿瘤患者生存率的降低。乏氧诱导因子=1(HIF-1)通常在人类肿瘤中过表达,其转录活性受细胞氧浓度的精确调节,是乏氧时进入细胞核激活转录的信使,继而可导致血管内皮生长因子(VEGF)和其他已知的HIF-1靶基因的转录激活。乏氧靶向治疗涉及多个方面,抑制HIF-1的活性是其中重要的一个方面。
Hypoxia is a pathophysiologic component of many disorders, contributes to the malignant phenotype and aggressive tumour behaviour and is associated with increased metastasis and poor survival. Hypoxia inducible factor( HIF)- 1 is overexpressed in common human cancers ,it's transcriptional activity is precisely regulated by cellular 02 concentration. HIF-1 can be viewed as a messenger that is sent to the nucleus to activate transcriptional responses to hypoxia and then leads to the transcriptional activation of vascular endothelial growth factor(VEGF) and other several dozen known target genes. Hypoxia targeted therapy for tumor involves many aspects,inhibitting the activity of HIF-1 is the important one of the aspects.
