摘要
目的了解大鼠晶状体脂质过氧化及抗氧化能力随年龄增长的变化趋势。方法雄性SD大鼠60只,按月龄不同分为1月龄组、3月龄组、6月龄组、12月龄组、18月龄组、24月龄组共6组,每组10只;处死动物取出眼球,采用硫代巴比妥酸比色法与化学发光法检测各组大鼠晶状体组织中谷胱甘肽(glutathione hormone,GSH)、超氧化物岐化酶(superoxide dismutase,SOD)、丙二醛(malondialdehyde,MDA)含量。结果大鼠晶状体GSH含量1月龄组、3月龄组、6月龄组、12月龄组、18月龄组、24月龄组分别为(30.49±7.39)mg、(33.32±6.13)mg、(29.04±3.06)mg、(28.65±4.84)mg、(22.03±4.24)mg、(20.53±3.22)mg;各增龄组GSH含量经F检验P<0.01,GSH含量与月龄负相关(P<0.01);大鼠晶状体SOD含量1月龄组、3月龄组、6月龄组、12月龄组、18月龄组、24月龄组分别为(24.63±1.25)U、(25.39±1.64)U、(23.17±1.81)U、(23.24±3.11)U、(20.47±1.08)U、(19.11±2.97)U;各增龄组SOD含量经F检验P<0.01,SOD含量与月龄负相关(P<0.01)。大鼠晶状体MDA含量1月龄组、3月龄组、6月龄组、12月龄组、18月龄组、24月龄组分别为(49.63±4.38)nmol、(52.19±3.82)nmol、(53.09±6.45)nmol、(53.59±4.66)nmol、(59.80±4.23)nmol、(62.21±3.93)nmol;各增龄组MDA含量F检验P<0.01,MDA含量与月龄正相关(P<0.01)。结论大鼠晶状体GSH、SOD及MDA随年龄增长分别呈降低、降低、升高趋势,提示晶状体老化过程伴随晶状体脂质过氧化损伤及抗氧化能力下降。
Objective To investigate the change of lipid peroxidation and antioxidation in the lens of aging rats. Methods Sixty Sprague Dawley (SD) rats were divided into 6 groups, including 1 month group, 3 months group, 6 months group,12 months group, 18 mouths group,24 months group, 10 rats in each group. Then rats were executed and eyes extirpated. The thiobarbituric acid and chemical luminescence methods were used to detect glutathione hormone ( GSH), superoxide dismutase (SOD) and malondialdehyde ( MDA ) conrained in the lens of these rats. Results The lens GSH contents were (30.49 ± 7.39)mg, (33.32±6.13 ) mg, (29.04 ± 3.06)mg, (28.65 ± 4. 84) mg, (22.03 ± 4.24) mg,(20.53±3.22) mg in 1 month group,3,6,12,18,24 months group. The lens GSH contents and age were inverse correlation( P 〈 0.01 ). The lens SOD contents were (24.63 ± 1.25)U, (25.39 ± 1.64)U, (23.17 ± 1.81)U, (23.24 ±3. 11)U, (20.47 ± 1.08)U,(19.11±2.97) U in 1 month group,3,6,12,18,24 months group. The lens SOD contents and age were inverse correlation( P 〈0.01 ). The lens MDA contents were(49.63±4.38)nmol, (52.19 ± 3.82)nmol, (53.09 ± 6.45) nmol, (53.59 ± 4.66) nmol, (59.80 ±4.23)nmol, (62.21 ± 3.93)nmol in 1 month group,3, 6,12,18,24 months group. The lens MDA contents and age were positive correlation (P 〈 0. 01 ). Conclusion With aging, the contents of GSH and SOD decreased and MDA increased,which indicates that the aging procedure is related with lipid peroxidation damage and the descent of antioxidation ability.
