摘要
目的:探讨基质金属蛋白酶-9(MMP-9)和组织型基质金属蛋白酶抑止因子-1(TIMP-1)在持续性房颤发生和维持中的作用。方法:30只犬随机分为对照组和房颤组,建立犬快速肺静脉起搏持续性心房颤动模型。取组织标本,用RT-PCR和Western印记法分别检测各组MMP-9和TIMP-1mRNA和蛋白的表达情况。结果:与对照组相比,房颤组肺静脉肌袖组织MMP-9mRNA与蛋白质水平均增高(P均<0.05),TIMP-1mRNA与蛋白质表达水平均降低(P均<0.05)。结论:MMP-9/TIMP-1平衡失调可能是房颤时肺静脉肌袖细胞外基质重构的因素之一,参与了房颤的发生和维持。
Objective: To investigate the effects of MMP - 9/TIMP - 1 (matrix metalloproteinases - 9/tiasue inhibitor of metalloproteinase - 1) on persistent atrial fibrillation(AF). Methods: Thirty dogs were randomly divided into two groups; normal control group(SR, n= 15) and atrial fibrillation group(AF, n= 15). The model of persistent atrial fibrillation was established by rapid pulmonary vein pacing in dogs. The blood samples were collected from left upper pulmonary veins of these animals. The mRNA and protein expressions of MMP - 9 and TIMP - 1 were assessed by RT - PCR and Western blot, respectively. Results:Compared with the control group, the mRNA and protein expressions of MMP - 9 increased significantly(P〈0. 05) and the mRNA and protein expressions of TIMP - 1 decreased dramatically(P〈0. 05) in AF group. Conclusion: The altered expression of MMP - 9 and TIMP - 1 contribute to extracelluar matrix remodeling in pulmonary vein myocardial sleeves and the remodeling is involved in the maintenance and stabilization of AF.
