Ki-67、p16和p53在结肠癌中的表达和相关性研究

摘要 目的:探讨Ki-67、p16和p53在结肠癌组织中表达的意义和它们之间的相关性。方法:应用链霉素-生物素(SP)免疫组织化学法,对127例结肠癌进行Ki-67、p16和p53的检测。结果:结肠癌Ki-67、p16和p53蛋白阳性表达率分别为74.8%、47.2%和54.3%。Ki-67蛋白阳性表达均与肿瘤分化程度、浸润深度和淋巴结转移显著相关(P<0.05),而与患者年龄、肿瘤大体类型无关(P>0.05);p16蛋白阳性表达与肿瘤分化程度和淋巴结转移显著相关(P<0.05),与患者年龄,肿瘤大体类型、浸润深度无关(P>0.05);p53与肿瘤分化程度显著相关(P<0.05),与患者年龄,肿瘤大体类型、浸润深度和淋巴结转移无关(P>0.05)。结肠癌中Ki-67蛋白和p16、p53蛋白的表达无显著关系(P>0.05)。结论:结肠癌中Ki-67表达的上调与其浸润转移密切相关,p16和p53在结肠癌发生发展过程中具有重要作用;术前联合检测Ki-67、p16和p53在结肠癌活检组织中的表达,对指导手术决策,以及估计预后有重要的临床应用价值。 Objective:To study the expression of Ki - 67, p16 and p53 in colon cancer tissues and their relationship. Methods:Using immunohistochernical method(SP method), the expression of Ki - 67, p16 and p53 was detected in 127 cases of colon carcinoma, Results: positive rates of Ki - 67, p16 and p53 were74. 8%, 47. 2% and 54. 3%, respectively. The positive expression of Ki - 67 was closely correlated to the differentiation degree,invasive degree and lymph ncde metastasis (P〉 05) , but it was not correlated to the age and organism type (P〉05);p16 was closely correlated to differentiation degree and lymph node metastasis(P〉0. 05), it was not correlated to the age, organism type and invasive degree (P〉0. 05); p53 was closely correlated to differentiation degree(P〉0. 05),it was not correlated to the age, organism type, invasive degree and lymph node metastasis There was no significant correlation among the expression of p53, p16 and p53 protein (P〉0. 05). Conclusion:Up regulation of Ki - 67 expression is closely correlated with invasion and metastasis of colon cancer, p16 and p53 play important roles in the development of colon carcinoma Detection of the expression of Ki - 67, p16 and p53 in biopsied tissues of colon cancer before operation is useful to guide the decision of operation and appraisal of its prognosis.
