生长激素对体外培养大鼠胰岛的促胰岛素释放功能的影响

摘要 【目的】探讨生长激素(GH)对体外培养大鼠胰岛的胰岛素释放功能的影响。【方法】对体外培养大鼠胰岛进行形态学观察及鉴定,分正常对照组及GH干预组,进行胰岛素含量测定及胰岛素释放试验。【结果】HG干预组胰岛培养6d后培养液中胰岛素总量显著高于正常对照组(P<0.05),在低糖及高糖条件刺激下的胰岛素释放量均显著高于正常对照组(P<0.05),两组间的胰岛素释放指数无统计学差异。【结论】GH对体外培养大鼠胰岛的胰岛素释放具促进作用,可能并不会因此加速胰岛功能的衰竭。为进一步探讨GH促进永生化后回复的胰岛细胞功能的恢复,提供了前期实验基础。 [Objective]To elucidate the effect of growth hormone(GH) on insulin secretion of rat islets cultured in vitro. [Methods]Rat islets were cultured in vitro and characterized. The gross insulin after 6-day cultivation and glucose-stimulated insulin secretion were determined in the control group and the GH group. [Results]The gross insulin in the medium from the GH group was higher than that of the control group ( P〈0.05). The secretion of insulin from the GH group in response to low glucose as well as high glucose was higher than that from the control group ( P 〈0.05), And there was no difference in insulin stimulation index between the two groups, [Conclusion]GH promotes the insulin secretion of rat islets cultured in vitro without accelerating the islets function failure. It indicates that GH may potentially help recover the insulin secretion function of the reversible immortalized β-cells.
