摘要
【目的】探讨免疫磁珠分选CD34+细胞及脐血中CD34+细胞含量的意义。【方法】脐血中分离单核细胞后,采用EasySep正选人CD34+免疫磁珠分选CD34+细胞,流式细胞仪检测分选前后CD34细胞的纯度。【结果】分选前MNC中CD34+细胞纯度为(1.14±0.71)%,分选后CD34+细胞纯度达到(91.55±4.11)%。【结论】脐血作为CD34+造血干/祖细胞的来源以及免疫磁珠方法在获得高纯度CD34+细胞中有重要的意义。
[Objective]To investigate the significance of immunomagnetic separation of CD34^+ cells from cord blood. [Methods] After mononuclear cells were isolated from umbilical cord blood, CD34^+ cells were separated from mononuclear cells by immunomagnetic cell sorting and their percentage was assessed by flow cytometry. [Results]The CD34^+ percentage of mononuclear cells before separation was (1. 14 ± 0. 71)%, while the CD34^+ percentage after immunomagnetic cell sorting was (91.55±4.11)%. [Conclusion]Both umbilical cord blood as a resource of CD34^+ hematopoietic stem cell/progenitor cell and immunomagnetic cell sorting play an important role in procuring CD34^+ cells with high purity.
