奥曲肽预适应对兔肝脏热缺血再灌注损伤的保护作用 被引量:1

Effect of Octreotide on Hemodynamics,Blood Biochemics and Inflammatory Cytokines in Hepatic Ischemia-reperfusion Injury

摘要 【目的】探讨奥曲肽对兔肝脏热缺血再灌注损伤的保护作用。【方法】采用兔肝脏热缺血再灌注模型,将24只新西兰大白兔随机分为3组:假手术组(A组)、肝脏缺血再灌注组(B组)和奥曲肽预适应组(C组)。观察三组动物缺血30min、再灌120min后血清丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)、乳酸脱氢酶(LDH)、脂质过氧化及血清中血浆内皮素(ET-1)的变化。【结果】肝脏缺血再灌注损伤时,血清ALT、AST、LDH、血浆ET-1、肝组织丙二醛(MDA)水平均显著增高,肝组织超氧化物歧化酶(SOD)明显减少。再灌注前使用奥曲肽可以明显减轻上述改变。【结论】奥曲肽可能通过保护肝细胞、下调肝脏内皮素的表达、抗脂质过氧化,从而减轻热缺血再灌注对肝脏的损伤作用。 [Objective]To explore the. protective effect of octreotide preeonditioning on liver isehemiareperfusion injury , [Methods] Pringle's maneuver liver ischemia-reperfusion models were established. Twenty four New Zealand Rabbits were randomly divided into three groups sham operation group (A group, n =8), ischemia-reperfusion group (B group, n=8) and octreotide preconditioning group(C group, n =8 ).In every group, the concentrations of serum ALT. AST. LDH. and the plasma level of endothelin-1 were measured 120min after reperfusion. [Results] After 120 minutes of reperfusion following 30 minutes of partial hepatic is chemia, the serum levels of ALT. AST and LDH and the plasma level of endothelin-1 increased significantly significantly. The hepatic-concentration of MDA showed an evident increase while the activities of SOD showed a markedly decrease. Octreotide pretreatment prevented hepatic warm ischemia reperfusion.injury.significantly [Conclusion](1)Otreotide shows its protective effects on the hepatic ischemia-reperfusion injury. (2) The protection, of octreotide on hepatic ischemia-reperfusion injury is related to the protective .effect for hepatic cells and de, pressing the overproduction of endothelin-1 and resisting lipid peroxidation.
