芳香开窍类中药含药脑脊液对星形胶质细胞水通道-4表达的影响

The influence of cerebrospinal fluid contained drugs of resuscitation with aromatics to the expression of aquaporin-4 mRNA of brain astrocyte

目的探讨芳香开窍类中药在治疗中枢神经疾病时的作用机制。方法原代培养小鼠大脑星形胶质细胞表达水通道-4mRNA,观察家兔含药脑脊液对原代培养的小鼠大脑星形胶质细胞水通道基因表达的影响。结果培养液中加入家兔脑脊液对水通道-4 mRNA表达没有影响(P>0.05),含石菖蒲的含药脑脊液使原代培养的小鼠大脑星形胶质细胞水通道-4基因表达降低(P<0.05);而含麝香的含药脑脊液对原代培养的小鼠大脑星形胶质细胞水通道-4基因表达无明显影响(P>0.05)。结论芳香开窍药石菖蒲可能能降低星形胶质细胞水通道-4基因的表达。

Objective It is to approach the mechanisms of some Chinese drugs for resuscitation with aromatics on treating central nerve disease. Methods The brain astrocytes （AST） express aquaporin- 4 （AQP4） mRNA was primary cultured. The effect of rabbit cerebrospinal fluid contained drugs on the expression of AQP4 mRNA of primary cultured brain AST of mice was observed. Results There was no influence on AQP4 mRNA when add rabbit cerebrospinal fluid to culture medium （P 〉 0.05）, The cerebrospinal fluid contained Grassleaf Sweetflag Rhizome depressed the expression of AQP4 mRNA （P 〈 0, 05） ; however the cerebrospinal fluid contained moschus had no remarkable effect on AQP4 mRNA （ P 〉 0.05）. Conclusion The Grassleaf Sweetflag Rhizome may reduce the expression of AQP4 mRNA of primary cultured brain AST of mice.
