FFA对足细胞活力的影响及丹酚酸黄芪多糖的干预作用研究 被引量:5

摘要 目的考察游离脂肪酸(FFA)对培养小鼠足细胞株活力的影响,以及丹酚酸与黄芪多糖的干预效果。方法取分化的足细胞,给予不同浓度(7.5μmol/L~480μmol/L)的游离脂肪酸(FFA)干预,观察其对足细胞活力(XTT)的影响,以确定FFA干预的浓度,并根据不同浓度丹酚酸与黄芪多糖对细胞活力的影响,确定丹酚酸与黄芪多糖的治疗浓度范围。根据观察结果,取适当的FFA干预浓度(30μmol/L),观察不同浓度丹酚酸(2.5mg/L~80mg/L)与黄芪多糖(0.05g/L~1.6g/L)对足细胞活力的保护作用。结果FFA自15μmol/L浓度开始,至480μmol/L,对足细胞活力有明显影响,使足细胞活力明显降低。丹酚酸2.5mg/L~80mg/L与黄芪多糖0.05g/L~1.6g/L对正常培养足细胞活力无明显影响,但对FFA30μmol/L浓度干预后的足细胞活力有明显的保护作用,其中丹酚酸在5mg/L~10mg/L浓度之间还呈现出剂量依赖关系,以10mg/L浓度作用最强,与对照比较差异有统计学意义(P<0.01),但随着浓度的增加,保护作用无明显增强。黄芪多糖0.1g/L~0.8g/L的浓度足细胞保护作用无明显差异。结论游离脂肪酸可明显降低培养足细胞的活力,丹酚酸与黄芪多糖有较好的保护足细胞活力作用,其中以丹酚酸10mg/L浓度作用最强。 Objective To observe the effect of free fat acid(FFA) on podocyte viability and its intervention by salvianolic acid(SAL) and Astragalus Polysaccharides (APS). Methods Viability of differentiated podocytes treated with palmitic acid (7.5 μmol/L-480 μmol/L), Sal or APS was measured to determine the suitable concentration of interventions. Then the protective effect of SAL (2.5 mg/L-80 mg/L) on FFA (30 μmol/L) treated with podocyte viability was compared with the effect of APS(0.05 g/l-1.6 g/L). Results FFA with different concentrations(15 μmol/L- 480 μmol/L) all could suppress podocyte viability. SAL(2.5 mg/L-80 mg/L) and APS(O.05 g/L-1.6 g/L) didn't affect normally cultured podocytes while they could protect the viability of podocytes treated with 30 p, mol/L FFA. There was a dose-effect relationship during 5 mg/L to 10 mg/L SAL while 10 mg/L SAL had the strongest effect (P〈O.O1). The effect didn't increase when the concentrations increase from 10 mg/L. Alas had similar effect during concentrations of 0.1 g/L to 0.8 g/L. Conclusion FFA could suppress viability of cultured podocytes. SAL and APS could protect podocyte viability, especially 10mg/L SAL.
