韧黏素在大鼠胚胎心脏发育过程中的表达 被引量:1

摘要 目的检测韧黏素-C(TN-C)和韧黏素-X(TN-X)在大鼠胚胎心脏中的表达,探讨其在胚胎心脏发育过程中的作用。方法雌性SD大鼠64只,雄性SD大鼠20只,雌雄同笼,以观察到雌鼠排出阴栓为妊娠0.5d(ED0.5),分别于ED12.5、ED15.5、ED19.5处死孕鼠,取出心脏。半定量反转录酶聚合酶链反应(RT-PCR)检测上述各孕龄大鼠胚胎心脏中TN-C和TN-X mRNA表达;免疫组织化学染色法观察上述各孕龄大鼠心脏TN-C和TN-X蛋白表达。结果TN-C mRNA在ED12.5大鼠胚胎心脏中高表达(0.6837±0.0431),ED15.5(0.4543±0.0388)和ED19.5(0.2505±0.0384)大鼠胚胎心脏TN-C mRNA表达下降,与ED12.5比较有显著性差异(Pa<0.05);ED12.5、ED15.5、ED19.5大鼠心脏TN-X mRNA相对表达量分别为0.7471±0.0433、0.7584±0.070、0.7671±0.088,各组间表达无显著性差异。免疫组织化学显示TN-C在ED12.5大鼠胎心有表达,在ED15.5少量表达,在ED19.5大鼠心脏未见表达;TN-X在ED12.5大鼠胎心的心室肌的细胞中有表达,在ED15.5、ED19.5胎鼠心脏中表达于整个心脏的心肌及房室间隔。结论TN-C在胚胎心脏早期发育过程中可能占有重要作用,而TN-X可能是心脏存活所必须的。 Objective To detect the expression of tenasein - C ( TN - C ) and tenascin - X ( TN - X) in the embryonic heart of rats, investigate their role during the heart development. Methods Sixty - four Spraque - Dawley female rats and 20 male rats were put into the same cage, when vaginal suppository was found , the femate was pregnant 0. 5 day ( ED 0. 5 ) , then executed the pregnant rats on ED 12.5, ED 15.5 and ED 19.5 ,got out the hearts. Semi - quantitative reverse transcriptase - polymerase chain reaction( RT - PCR) was used to detect the TN- C and TN- X mRNA expression in the rat hearts aged embryonic day 12.5 (ED 12.5 ) ,ED 15.5 and ED 19.5, The protein expression of TN - C and TN - X in the embryonic hearts was detected by immunohistochemistry. Results TN - C mRNA was highly expressed in ED 12.5 embryonic hearts(0. 683 7 ±0. 043 1 ), the expression of TN - C mRNA of ED 15.5 (0. 454 3 + 0. 038 8 ) and ED 19.5 (0. 250 5 ± 0. 038 4 ) embryonic hearts decieased ,they were different from the expression of ED 12.5 ( Pa 〈 0. 05 ). The expression of TN - X mRNA of the embryonic hearts of ED 12.5, ED 15.5, ED 19.5 was 0. 747 1 ± 0. 043 3,0.758 4 ±0. 070 and 0. 767 1 ± 0. 088, respectively. There was no difference between the groups. Immunohistochemistry proved that TN - C protein was present in embryonic rat heart of ED 12.5, was poorly in heart of ED 15.5 ,and absent in heart of ED 19.5 ;Also it proved that TN - X was present in the myocardium of ventricle in hearts of ED 12.5,in hearts of ED 15.5 and ED 19.5. TN - X was expressed in the whole heart. Conclusion TN - C may play an important role in the early heart development, and TN - X may be necessary for the survival of heart.
