摘要
目的:探讨重组人IL1-Ra与TGF-β1基因转染软骨细胞的表达情况。方法:体外分离培养兔关节软骨细胞,然后用构建的IL1-Ra与TGF-β1质粒通过脂质体介导下共转染软骨细胞,通过荧光显微镜观察转基因的表达和荧光定量PCR检测其表达。结果:荧光显微镜观察有绿色荧光蛋白表达,荧光定量PCR鉴定证实转染的软骨细胞中转基因得到表达。结论:IL1-Ra与TGF-β1基因共转染软骨细胞可以获得表达,为基因治疗骨关节炎的研究提供基础。
Objective: To study the expression of recombined human IL1-Ra and TGF-β1 genes after cotransfecting them into articular chondrocytes of rabbits in vitro. Methods: Articular chondrocytes of rabbit were isolated and cultured in vitro. The plasmids carrying IL1-Ra and TGF-β1 genes were cotransfected into chondrocytes by liposome method. The expression of them was observed under a fluorescence microscope and detected using TaqMan Real-Time PCR assay. ReCta: Enhanced green fluorescent protein was observed. Real-Time PCR analysis showed that the genes were expressed in chondrocytes. Conclusions: IL1-Ra and TGF-β1 genes could be expressed effectively after being cotransfected into chondrocytes, which provides a basis for gene therapy ofosteoarthritis.
