摘要
目的建立大鼠冠状动脉结扎后急性实验性心肌缺血模型及观察益心宁对其的影响。方法在大鼠肺动脉圆锥与左心耳根部下方约2mm处结扎冠脉左室支,造成大鼠急性心肌缺血,将其随机分为益心宁8g、4g、2g.kg-1组及阳性对照愈风宁心片2g.kg-1组,另设模型对照组和正常对照组,1次性灌胃给药,记录给药后4、5、6h的胸前多导联心电图,末次记录完心电图后,从大鼠颈静脉取血2mL,测定血清肌酸激酶(CPK)及肌酸激酶同功酶(CK-MB)活性,并计算缺血面积占心室面积的百分比。结果益心宁8、4、2g.kg-1一次性灌胃给药,能缩小大鼠心肌缺血面积,降低大鼠血清肌酸激酶(CPK)及肌酸激酶同功酶(CK-MB)活性,使升高的∑ST明显降低。结论益心宁对大鼠心肌缺血具有明显的保护作用。
OBJECTIVE To study the effect of Yixinning (YXN) on acute myocardial ischemia in rats. METHODS Acute myocardical ischemia models were established by ligating the aeteria coronaria of rats and these rats were randomly divided into YXN high dose group (8g · kg ^-1), YXN medium dose group (4g · kg^-1 ),YXN low dose group (2g · kg^-1),positive control group (YFN tablet, 8g · kg^-1 ), control group and model control group. After intragastric administrated of YXN 4,5,6 hours,the epicardial ECG were recorded,and the aetivities of serum CPK,CK-MB and percentage of the myocardial ischemia area in ventricles were evaluated. RESULTS Compared with the model control group,treatment with YXN could reduce the MI area and the activities of serum CPK and CK-MB, meanwhile ∑ST was significantly decreased in YXN group. CONCLUSION YXN had significant protective effect on acute myocardial ischemia in rats.
