摘要 目的建立并评价检测肠杆菌科细菌氦苄西林头孢菌素β-内酰胺酶(AmpC酶)的乙二胺四乙酸(EDTA)-头孢西丁(FOX)平板试验(E-CAM)。方法应用E-CAM和酶粗提物三维试验(TDEM)同时检测临床分离的FOX耐药肠杆菌科细菌的AmpC酶,对2种方法进行比较,并以阴沟肠杆菌(029M)和大肠埃希菌(ATCC 25922)作质控。结果E-CAM试验应用6μg/mL FOX平板和750μg EDTA进行检测,其结果与TDEM完全一致。结论该方法操作简便,结果易于判读。在一块平板上可同时进行多株菌的检测,适于各级临床微生物实验室应用。 Objective To set up and evaluate a new ethylenediamine tetra-acetic acid (EDTA)-cefoxitin (FOX)- agar medium (E-CAM) for detecting ampicillin cephamycin β-1actamases (AmpC) in Enterobacteriaceae. Methods The E-CAM method was compared to the previously published three-dimensional extract method (TDEM) for the detection of AmpC. Clinical isolates of FOX-resistant Enterobacteriaceae were tested by both methods. Positive control (Enterobacter cloacae 029M) and negative control ( Escherichia coli ATCC 25922) strains were included on each plate. Results The E-CAM method with 6 μg/mL of FOX and 750 μg of EDTA were similar to the TDEM method for detecting AmpC production in Enterobaeteriaceae, the coincidence rate was 100%. Conclusions The new method is easier to perform and interpret and allows for testing muhiple isolates on a single plate. Clinical laboratories can use this technique to detect the presence of AmpC enzyme.
