摘要
利用CA125抗原免疫Balb／c小鼠，采用杂交瘤细胞技术，建立分泌抗CA125抗体的杂交瘤细胞株。将CA125单克隆抗体应用到化学发光免疫分析和免疫放射分析方法中，病理值的测定结果和进口试剂盒测定值相关方程分别为y=0．85x＋11．1，r=0．932和y=0．87x-12．8，r=0．983；正常血样测量值范围分别为3．3～26．7和1．5～27．0U／mL。CA125单克隆抗体制备的包被板和包被管在37℃下可稳定存放14d，且不影响分析结果。
Balb/c mice were immunized with carbohydrate antigen 125 （CA125）, and a competing sandwich immunoassay was used to screen positive clone of hybridoma cell, finally one monoclonal antibody against CA125 was got successfully. Applications of the antibody in immunoradiometric assay （IRMA） and chemiluminescence immunoassay （CLIA） show it is an ideal substitute of coating part in the sandwich system. We performed a normal range studies in which all healthy women had assay values less than 35 U/mL. A direct relationship exits between ELIA and IRMA with commercial kits, correlative equations are y=0.85x＋11.1 （r=0. 932） in CLIA, y=0.87x-12.8（r= 0. 983） in IRMA, respectively. New antibody is stable at 37℃ for 2 weeks.
