The expression of dopamine transporter in aged rat brains

摘要 目的观察老年大鼠脑内多巴胺转运体(DAT)的表达变化。方法健康雄性Wistar大鼠3月龄、12月龄和24月龄组各20只;应用免疫组织化学和免疫印迹技术观察大鼠脑内多巴胺转运体在衰老过程中的表达。结果大鼠黑质、尾壳核多巴胺转运体免疫反应强度在24月龄组明显低于3月龄(P<0.01)及12月龄组(P<0.05);24月龄组大鼠黑质、尾壳核DAT与β-actin免疫印迹条带相对吸光度的比值低于3月龄和12月龄组(P<0.05,P<0.01)。结论大鼠脑内多巴胺转运体的表达随衰老而减少。 Objective To study the altered expression of dopamine transporter (DAT) in aged rat brains. Methods The healthy male Wistar rats were divided into 3-, 12- and 24-month-old groups (n = 20). Immunohistochemistry and Western-blot were used to detect the altered expression of DAT protein in aged rat brains. Results DAT immunoreactive intensity in substantia nigra and caudate putamen in 24-month-old group was lower than that in 3-month-old (P 〈0. 01 ) and 12-month-old (P 〈0. 05) groups. The ratio of DAT/13-actin in substantia nigra or caudate putamen of 24-month-old rats was significantly lower than that in 3- and 12-month-old rats ( P 〈 0. 05, P 〈 0. 01 ). Conclusions The expression of DAT in aged rat brains decreases with aging.
