摘要
目的观察复方刺五加注射液(CASI)对大鼠心肌缺血再灌注(I/R)损伤的保护作用及其机制。方法通过结扎大鼠冠状动脉左前降支30min,再灌注120min,建立心肌I/R损伤模型,计算心肌梗死范围(MIS),测定血清天门冬氨酸氨基转换酶(AST)、磷酸肌酸激酶(CK)、乳酸脱氢酶(LDH)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性及丙二醛(MDA)含量,血浆前列环素(PGI2)及血栓素A2(TXA2)水平。结果CASI40、80mg/kg对心肌I/R损伤大鼠,可明显缩小MIS,降低血清AST、CK、LDH活性及MDA含量,提高SOD及GSH-Px活性,使血浆TXA2水平明显下降,PGI2水平及PGI2/TXA2比值明显增高。结论CASI对大鼠心肌I/R损伤具有明显保护作用,可能与其减少自由基对心肌的氧化损伤,增强抗氧化酶活性,纠正PGI2/TXA2失衡等机制有关。
Objective To investigate protective effect of compound acanthopanax senticosus injection (CASI) on myocardial ischemia-reperfusion (I/R) injury in rats and its mechanism. Methods The myocardial I/R model was induced by left anterior descending coronary occulusion in 30 min and reperfusion in 120 min in rats. The changes of myocardial infarct size (MIS), serum aspartate aminotransferase (AST) , creatine phosphokinase (CK) and lactate dehydrogenase (LDH) activity, malondialdehyde (MDA) content, superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) activity, and plasma prostacycline ( PGI2 ), thromboxane A2 (TXA2 ) levels were determined..Results 40, 80 mg/kg CASI significantly reduced MIS, reduced serum AST, CK, LDH activity and MDA content, and increased SOD and GSH-Px activity, declined plasma TXA2 level, enhanced plasma PGI2 level and PGI2/TXA2. Conclusions CASI has protective effect on myocardial I/R injury in rats by reducing oxidative damage of free radicals on myocardium, enhancing activity of antioxidase and correcting disequilibrium of PGI2/TXA2 etc.
