摘要
建立了HPLC法测定截短侧耳素的含量,采用Agilent Extend C8色谱柱,流动相为乙腈-甲醇-0.02mol·L^-1的磷酸二氢钾缓冲液(35:10:55,体积比),流速为1.00mL·min^-1,检测波长为205nm。截短侧耳素质量浓度在40~2400μg·mL^-1范围内线性关系良好(r^2=0.9999),定量限为0.08μg,检测限为0.04μg。
A HPLC method for the determination of pleuromutilin was developed. The sample was separated on a Agilent Extend Cs eluted at a flow rate of 1.00 mL · min^-1 with mobile phase of acetonitrile-methanol-phosphate buffer (35 : 10 : 55,V/ V), pleuromutilin was detected and quantified by the absorbance at 205 nm. The linearity was obtained over the range of 40~ 2 400 μg · mL^-1 for pleurornutilin(r^2 =0. 999 9). The quantitative limit is 0.08 μg,and the detection limit is 0.04 μg.
