截短侧耳素的HPLC测定

Determination of pleuromutilin by HPLC

建立了HPLC法测定截短侧耳素的含量，采用Agilent Extend C8色谱柱，流动相为乙腈-甲醇-0．02mol·L^-1的磷酸二氢钾缓冲液（35：10：55，体积比），流速为1．00mL·min^-1，检测波长为205nm。截短侧耳素质量浓度在40～2400μg·mL^-1范围内线性关系良好（r^2=0．9999），定量限为0．08μg，检测限为0．04μg。

A HPLC method for the determination of pleuromutilin was developed. The sample was separated on a Agilent Extend Cs eluted at a flow rate of 1.00 mL · min^-1 with mobile phase of acetonitrile-methanol-phosphate buffer （35 ： 10 ： 55,V/ V）, pleuromutilin was detected and quantified by the absorbance at 205 nm. The linearity was obtained over the range of 40～ 2 400 μg · mL^-1 for pleurornutilin（r^2 =0. 999 9）. The quantitative limit is 0.08 μg,and the detection limit is 0.04 μg.
