摘要
利用RT-PCR技术,从纽荷尔脐橙(Citrus sinensis Osbeck)果实cDNA中分离出类胡萝卜素异构酶(Carotenoid isomerase,CRTISO)基因片段。该片断长446bp,编码148个氨基酸。用BLASF比较其氨基酸同源性发现,该片段氨基酸残基与伏令夏橙(C.sinensis Osbeck)、番茄(Lycopersicon esculentum Miller)和拟南芥(Arabidopsis thaliana)CRTISO的同源性分别为99%、85%和83%,与里斯本柠檬(C.limon Burm.F.)和温州蜜柑(C.unshiu Marc.)完全一致。半定量RT-PCR分析表明,CRTISO基因在8~11月份纽荷尔果实中均有表达,10月份的表达量最大。
The cDNA sequence for carotenoid isomerase (CRTISO) has been cloned from fruits of Newhall navel orange by RT- PCR technique. Sequencing results revealed that it shared the length for 446bp, encoding 148 amino acids. BLAST results showed that the deduced amino acid shared 100% identities with those of Lisbon limon and Satsuma Mandarin, 99% identities with Valencia sweet orange CRTISO, 85% and 83 % identities with those of Lycopersicon esculentum Miller and Arabidopsis thaliana, respectively. Semi-quantitative RT- PCR results showed that expressions of CRTISO gene in fruits of Newhall navel orange were detected from August to November, with the highest one in October.
