人白细胞介素-6与肿瘤坏死因子突变体融合蛋白对人骨髓瘤细胞的细胞毒作用 被引量:4

SCytotoxic activity of a human interleukin 6 and mutative tumor necrosis factor fusion protein of human myeloma cells

摘要 本文报道了一种高活性人肿瘤坏死因子(TNF)突变体与人白细胞介素-6(IL-6)的融合蛋白在大肠杆菌中表达产物的纯化及其对L929细胞和人骨髓瘤细胞的细胞毒作用。结果表明,该融合蛋白既具有TNF抗肿瘤活性,又具有对高表达IL-6受体的肿瘤细胞特异性的杀伤作用。 The purification and cytotoxicity of a 5′IL6 TNFΔ,highly expressed in E.coli ,were studied.The fusion protein was purified by gelfiltration.5′IL6 TNFΔ could kill both L929 and human myeloma cells.Blocking the IL 6 receptors on myeloma cells decreased the cytotoxicity by 30 times.The antitumor activity almost disappeared as the mutant protein was blocked by anti TNF antibodies.The activity of TNF mutant was 3 times higher than that of the fusion protein.It was suggested that TNF be the anti tumor domain.The fusion of IL6 could improve the recognization of 5′IL6 TNFΔ to tumor cells especially with large amount of IL 6 receptors,but the configuration of TNF affected.
