摘要
应用组蛋白H1激酶活性分析技术,检测视黄酶和芳维甲诱导HL60细胞分化过程中细胞周期素依赖的蛋白激酶CDKs活性变化。结果显示处理3~4天的细胞大部分已向粒系分化,CDK2和CDK4激酶活性显著降低。推测细胞周期素依赖的蛋白激酶活性下降可能与视黄酸类物质诱导HL60细胞分化有关。
The role of cyclin dependent kinases in differentiation of HL 60 cells has been studied by analysing the changes of cyclin E and cyclin D 1 associated kinase activities by histone H 1 kinase assays.During differentiation induction with retinoic acid and arotinoid ethylester,both cyclin E and cyclin D 1 associated kinase activities were decreased,which may partially owing to upregulation of CDK inhibitors(CKIs) that could bind and inactivate cyclin/CKD complexes.Our data suggest that inhibition of CDK activity may be relative to granulocytic differentiation of HL 60 cells.
